Cytokines and chemokines as therapeutic targets for ischemic kidney injury
نویسندگان
چکیده
Cytokines and chemokines induce the migration, activation, proliferation, and/or differentiation of inflammatory cells and resident cells, and participate in the pathogenesis of some autoimmune diseases. Some of these molecules are effective therapeutic targets. Anti-tumor necrosis factor (TNF)-α and anti-interleukin (IL)-6 therapies are effective in patients with rheumatoid arthritis, and have markedly altered the therapeutic strategies for this disease. In ischemic kidney disease, cytokines and chemokines also play key roles in the progression of tissue destruction or remodeling. Accumulated data regarding ischemic kidney disease suggested that chemokines or chemokine receptors participate in unique pathological changes at particular time points in renal injury. Some molecules involved in these cytokine/chemokine cascades are potential therapeutic targets. In addition to antagonists for single chemokine receptors, promiscuous antagonists for chemokine receptors are currently under investigation. Further studies are needed to develop cytokineand chemokine-based therapeutic strategies for ischemic kidney injury. Rec.3/15/2011, Acc.4/8/2011, pp365-369

Involvement of inflammation in autoinflammation and autoimmune disease
Various recent studies reveal that many autoinflammatory syndromes result from inflammasome related protein abnormality. Some inflammasomes closely correlate with caspase 1. Caspase 1 is a key molecule for activation of IL-1β, which is responsible for induction and augmentation of inflammation. Inflammatory cytokines, including IL-1β, are necessary for TH17 differentiation from TH0 cell. TH17 c...
متن کاملTranscriptional analysis of infiltrating T cells in kidney ischemia-reperfusion injury reveals a pathophysiological role for CCR5.
Although T cells have been shown to play a direct role in kidney ischemia-reperfusion injury (IRI), little is known about the underlying mechanisms. We hypothesized that studying the transcriptional responses in kidney-infiltrating T cells would help elucidate novel therapeutic targets for kidney IRI. Unilateral renal pedicle clamping for 45 min was performed in male C57BL/6 mice, and CD3(+) T ...

Objective(s): Renal ischemia-reperfusion injury (IRI) as a severe condition of acute kidney injury (AKI) is the most common clinical problem with high mortality rates of 35-60% deaths in hospital. Mesenchymal stem cells (MSC) due to unique regenerative characteristics are ideal candidates for the treatment of the ischemic injuries. This work is focused on the administration of MSC to IRI-induce...
متن کاملEstrogen and P2 Purinergic Receptor Systems in Microglia: Therapeutic Targets for Neuroprotection.
Microglia, the primary resident immune cell population in the CNS, react to signals of injury or infection and produce inflammatory cytokines, chemokines, and reactive oxygen species, many of which can be neurotoxic in large quantities. Indeed microglial hyperactivation is thought to contribute to the pathology of many neurodegenerative disorders as well as ischemic and traumatic brain injuries...
متن کاملThe role of chemokines and chemokine receptors in mycosis fungoides
Background: Chemokines are a family of cytokines initially characterized by their capacity to induce chemotaxis, or directed leukocyte migrations. These receptors are activated by chemotactic cytokines called chemokines. Interactions between chemokines and chemokine receptors also are involved in tumorigenesis, migration and invasion of lymphoma cells. Methods: An English literature search...
